چکیده

Seedlings of Arabidopsis thaliana, Columbia ecotype, were used in vitro to analyze root hair growth response to inoculation by the plant-growth-promoting rhizobacterium, Azospirillum brasilense Sp-245. Root hair length was measured at physiologically-identical stages of root growth. In seedlings inoculated with A. brasilense, root hairs were more than twice the length of those of the non-inoculated control. This effect was consistent, reproducible and independent of N or C sources in the growth medium (although root growth was significantly suppressed in C-starved seedlings). Root hair growth promotion was similar in media with or without 1 % sucrose. Although root hair length of non-inoculated seedlings growing in the presence of various amounts of potassium nitrate (0-20 mM) was affected, it was significantly lower than that caused by inoculation with A. brasilense. We propose that A. brasilense inoculation affects the size of individual root hairs of Arabidopsis plants in vitro.

Rhizobacteria for reduced fertilizer inputs in wheat (Triticum aestivum spp. vulgare) and barley (Hordeum vulgare) on Aridisols in Turkey

The present study assessed the effect of seed inoculation with single or multiple plant growth-promoting rhizobacterial (PGPR) strains on yield in spring wheat (Triticum aestivum spp. vulgare var. Kırık) and barley (Hordeum vulgare cv. Tokak) plants in both greenhouse and field conditions during the years 2007 and 2008. The treatments of wheat and barley plants during the first year include...

Effect of Inoculation with Azospirillum brasilense Strains on the Germination and Seedlings Growth of the Giant Columnar Cardon Cactus (Pachycereus pringlei)

Wild Cardon cactus seeds were inoculated with Azospirillum brasilense strains Cd and Sp-245 to improve seed germination and seedling growth parameters. Differential germination responses were related to the bacterial strain. A. brasilense Cd (the type strain for the species A. brasilense) significantly decreased seed germination. A. brasilense Sp245 (a known beneficial bacteria for cereal plant...

Enhanced Growth of Wheat and Soybean Plants Inoculated with Azospirillum brasilense Is Not Necessarily Due to General Enhancement of Mineral Uptake.

The capacity of Azospirillum brasilense to enhance the accumulation of K, P, Ca, Mg, S, Na, Mn, Fe, B, Cu, and Zn in inoculated wheat and soybean plants was evaluated by using two different analytical methods with five A. brasilense strains originating from four distinct geographical regions. A Pseudomonas isolate from the rhizosphere of Zea mays seedlings was included as a control. All A. bras...

Analysis of SFL1 and SFL2 Promoter Region in Arabidipsis thaliana using Gateway Cloning System

SFL1 and SFl2 (SETH Four Like) genes are two members of SETH4 gene family in Arabidopsis thaliana expressed in saprophytic tissues. In this study, expression of SFL1 and SFL2 genes were studied using Gateway Cloning Technology. Primers were designed for PCR amplification of promoter region of SFL1 (900 bp) and SFL2 (930 bp) genes having attB1 recombination sites using Kod Hi Fi DNA polymerase e...

Effect of Azospirillum brasilense inoculation on rhizobacterial communities analyzed by denaturing gradient gel electrophoresis and automated ribosomal intergenic spacer analysis

Nucleic acid-based techniques allow the exploration of microbial communities in the environments such as the rhizosphere. Azospirillum brasilense, a plant growth promoting rhizobacterium (PGPR), causes morphological changes in the plant root system. These changes in root physiology may indirectly affect the microbial diversity of the rhizosphere. In this study, the changes in the rhizobacterial...
